Experiment: Creating a Liquid Culture Jar Using Self-Healing Injection Ports

Materials:

  • Sterilized liquid culture jar with nutrient solution
  • Self-Healing Injection Port (affixed to jar lid)
  • Sterile syringe with Luer Lock End Cap
  • Mushroom spore syringe or agar wedge
  • Tinfoil
  • Pressure cooker or autoclave
  • Micropore tape
  • Sterile gloves

Procedure:

1. Prepare the Liquid Culture Jar:

  • Cover the top half of the jar with tinfoil so as to cover the lid completely. Sterilize the jar containing the nutrient solution with the Self-Healing Injection Port using a pressure cooker at 15 PSI for 30 minutes. Ensure the injection port is securely fixed to the jar lid before Heat Sterilization.

2. Sterilization and Setup:

  • Working in a sterile space (Still air box or Flow hood)
  • Wait till the jar has cooled then remove the tinfoil covering the lid.
  • Prepare your spore syringe or an agar wedge.

3. Inoculate the Liquid Culture:

  • If using a spore syringe: Insert the syringe needle through the Self-Healing Injection Port and inject 1-2 mL of spore solution into the nutrient solution.
  • If using an agar wedge: Use a sterile tool to transfer a small agar wedge into the jar. You must be inside a sterile environment (like a Still-air box or Flow Hood ).

4. Seal and Incubate:

  • Once inoculated, remove the needle. The Injection Port will automatically seal itself.
  • Place a piece of micropore tape over the injection port to further secure it.
  • Incubate the jar at 25-27°C for 7-14 days, shaking it once daily to promote even mycelium growth in the liquid culture.

5. Harvesting Liquid Culture:

  • Once the jar is 50-80% colonized, the liquid culture is ready for use. Move into the sterile space and peel off the micropore tape to reveal a Sterile injection port. There is no need to Sterilize the injection port again as the micropore tape will keep the port clean and when removed will leave a clean surface. You can now extract the culture with a sterile syringe for further inoculation of grain spawn or substrate jars.

6. Testing the Liquid Culture:

  • For safety, always test the liquid culture by inoculating a small amount onto an agar dish to ensure it is contamination-free before mass inoculation.
