Experiment: Sealing and Sterilizing a Mushroom Cultivation Lid with High-Temperature Silicone Sealant
Materials Needed:
- High temperature silicone sealant
- Jar lid for mushroom cultivation (with pre-drilled hole)
- Alcohol swabs
- Sterile gloves
- 16mm syringe filter
- Masking tape
- Small spatula or applicator tool
Procedure:
- Prepare the Jar Lid: Sterilize the jar lid with alcohol swabs and wear sterile gloves thus limiting contamination.
- Make the Injection Port: Cover the hole in the jar lid with masking tape from the inside of the lid so no silicone spills through when applying the silicon. Using the high-temperature silicone sealant, carefully apply a thin layer around the pre-drilled hole on the outside of the lid. Ensure the sealant forms a clean, smooth surface. This seal will serve as an injection port once fully cured.
- Attach the Syringe Filter: Apply a small dab of the high-temperature silicone to the edge of the syringe filter. Place the filter on the lid to cover the hole completely, ensuring a tight seal. Let the silicone set for 24 hours to fully cure.
- Test the Seal: After 24 hours, check the seal by gently pressing the filter and ensuring no air escapes. Sterilize the jar and lid in a pressure cooker at 15 PSI for 60-90 minutes to test the durability of the seal.
- Usage: The sealed lid is now ready for use in mycology for sterile liquid cultures or substrate inoculation. You can safely inject spores or liquid culture through the silicone port using a sterilized syringe.
