Experiment to Grow Lion’s Mane Using Liquid Culture
Objective:
To successfully grow Lion’s Mane mushrooms using liquid culture inoculation into a hardwood sawdust substrate.
Materials Needed:
- Lion’s Mane liquid culture (Hericium erinaceus)
- Hardwood sawdust substrate
- Wheat bran (10-20%)
- Sterilized fruiting bags (autoclave filter grow bags)
- Still air box or flow hood
- Sterile syringe for inoculation
- Incubator @ temperature (20-26°C)
- Sharp knife for harvest
Procedure:
- Substrate Preparation:
Create a substrate mix using hardwood sawdust with 10-20% wheat bran. Sterilize the mixture in fruiting bags using an autoclave or pressure cooker. - Inoculation:
In sterile conditions (inside a still air box or flow hood), inject Lion’s Mane liquid culture into the sterile substrate bag using a sterile syringe. Shake the bag to evenly distribute the culture throughout the substrate. - Incubation:
Place the inoculated bag in an incubator set at 25-26°C. Allow it to incubate for 2-3 weeks, shaking the bag once a day to aid colonization. Observe the mycelium as it starts to grow through the substrate. - Fruiting Process:
Once the substrate is fully colonized, open the bag slightly by cutting small x’s in areas where primordia (fruiting bodies) have formed. Expose the bag to indirect light and maintain humidity at high levels. Do not spray water directly on the fruiting bodies. - Harvesting:
When the Lion’s Mane mushrooms reach maturity (2-3 weeks), harvest them by cutting them off at the base with a sharp knife. Handle the fruiting bodies gently to avoid breaking the delicate spines. - Subsequent Flushes:
Allow the bag to remain in the grow area to encourage additional flushes. Mushrooms will grow from previous sites.
